She was the eldest surviving child of Ulrik Frederik Gyldenløve and his third wife Antoinette Augusta von Aldenburg. She married her cousin Christian Gyldenløve, count of Danneskiold-Samsøe, in 1696. She gave birth to two children; Christiane Charlotte , who died before her second birthday and only three days after her sister Friederike Louise had been born. Charlotte Amalie died two months later.

Like her husband and mother in law she might have been transferred to St Petri Church in 1734.
